It was in the flashbacks. He was wheelchair bound while working the box firm and his coworkers made fun of him for wanting to go on the expedition. Also, when he got there, the tour manager wouldn't let him on the bus. He said he didn't anticipate the handicap and that it wasn't safe for him. Good thing he can walk now because the island isn't very ADA acceptable.

I also missed the first season. In the flashbacks from last night, it didn't show anything about Locke in a wheelchair. Whats the story?

I remember the episode that showed the flashback of Locke in a wheelchair-- that was a great episode. It was early in the first season, probably on disc 1 or 2.

We're still missing a part of Locke's story-- the part between last night's flashback with girlfriend Helen-- and the part where he somehow gets paralyzed.

didn't it show him getting hit by a car at some point?

You're confusing "Lost" with "My Name is Earl."

(I don't remember the referenced car scene.)
